What is PE-22-28?
PE-22-28 10mg is a seven amino acid peptide studied in mice. The two studies cited here come from one French team, in Valbonne, and none of the work cited involved people.
PE-22-28 is a trimmed-down form of spadin, a 17 amino acid peptide that the body cuts from a larger protein named sortilin. Both target TREK-1, a channel in the walls of nerve cells that lets potassium pass.
Shutting that channel is the idea behind the whole line of work. The researchers sit at the Institute of Molecular and Cellular Pharmacology in Valbonne, part of the French national research centre.
What the 3 PubMed papers cited for PE-22-28 looked at, 2010 to 2019
Spadin made its debut in 2010, when the team put mice given it through five standard behaviour tests used to screen antidepressant drugs.
Seven years later the same team built PE-22-28 out of spadin. They measured how firmly the shorter peptide shut TREK-1 and how long it lasted in the body, and ran it through the same mouse tests.
Looking back in 2019, a review gathered the work on spadin and its shortened forms in mouse tests, and noted that spadin itself breaks down within hours.
Tests such as the forced swim test are screening tools, used to sort compounds. On their own they say nothing about people.

Reading the PE-22-28 record
This record is tidy but narrow. The same French team designed the parent peptide, made the short version and ran the key tests cited here. That keeps the story clear, and it also means the experiments cited rest on one team's work.
The tests themselves are screening models in mice, built to sort compounds quickly. A mouse test of this kind is a reason for more study. It is not a finding about mood in people.
Two details round out the picture. Sortilin is the larger protein from which spadin is cut, and spadin breaks down within hours. The 2017 paper measured how long the shorter versions, PE-22-28 among them, last in the body.
Questions about PE-22-28
Has PE-22-28 been tested in people?
Not in any study our search found. The two experiments cited here used mice and came from one French team, in Valbonne.
What is TREK-1?
A channel in the wall of a nerve cell that lets potassium through. Spadin and PE-22-28 both block it.
Where does the name spadin come from?
Spadin is a piece cut from a protein called sortilin; PE-22-28 is a shorter, lab-made version of that piece.
